Hi Hermann,
thank you for your tips and questions. They brought us on the right
track!
We had to change a path setting in /etc/init.d/brltty (from
DAEMON=/sbin/brltty to DAEMON=/bin/brltty)
The brltty daemon now starts at boot time und both braille devices work
correctly.

Your syntax is for usb2serial.
To adjust your startup settings take a look at /etc/brltty.conf
What's teh result of
update-rc.d brltty defaults
Does the display start with the system?
What's the result of
cat /etc/default/brltty
