Is it possible to use a uri in the http://host/file form when using
GFile? I tried this in my code and it doesn't seem to work, so I'm
wondering what the next best alternative is for this.

Yes, that is one of the reasons GIO exists in the first place. HTTP
support is still being worked on for both Windows and *nix, but should
be in a somewhat usable state now. You may want to look over your code
and check for warning and/or errors that may be occurring to see what
isn't working. Some things just don't work yet, however, like setting
attributes on files and folders via GVFS on *nix.

Bugs should be reported under GIO when using HTTP on Windows, and
GVFS's HTTP-backend when using HTTP on Linux. Tor Lillqvist is working
on the Windows side of things, while Christian Kellner is working on
the HTTP support for GVFS. And you can always ask any questions on
GVFS support on the GVFS-list.
