Information is entered in Tracker either by "miners" or directly by
applications. The miners are small modules that crawl data sources
(for example, your filesystem) and insert data in Tracker. Currently
Tracker ships with two miners, one crawling the FS and another the
installed applications. So basically what you have to do is configure
the FS miner to ensure it's crawling the right dirs for you.

But this miner is going to add only the plain text contents of the
files, right? Or does it e.g. look in the extended attributes for
additional metadata?
Actually, the miner will enter basic metadata into Tracker (filename,
modification date etc.) and will search then for an extractor for that
file.
The extractor knows about the file format, and therefor extracts all the
metadata (for example title, duration, exif metadata...).
Extended attributes are not used AFAIK.

If you want to add custom triples in the store, you can use the
tracker-sparql tool.

Just to make sure: my metadata, keywords and timestamps would be such
additional triplets, right?
