Hello again,
I had forgot to build Python:
$ jhbuild build python
Building itstool is now ok, sorry for the noise.
Maybe, it would be still worth to mention it on wiki:

PS: by the way:
The shell script name gtk-osx-setup.sh and link is not correct in section Getting Started page:
