I think you MUST print the "INSTALL" file that comes with glib and gtk sources, and read it carefully.
It can be a good idea to remove the previous glib and gtk installed files before installing the new releases.
First:
configure glib --prefix=/usr/local
make
test it
make install
Then the same for gtk
