Knowledge of Xlib isn't absolutely necessary to use GTK. However, as
someone who started to learn Xlib/Xt and decided to switch to GTK, I have
found that knowing a little about Xlib makes it easier to decipher how
some of the not-so-well-documented functions in GTK/GDK work. For
example, the colormap and drawing related things are essentially
simplified wrappers for the equivalent Xlib functions.
