I do:
% echo $PATH
/usr/local/bin:/bin:/usr/bin:/usr/X11R6/bin (etc...)
When I run gtk-config from a arbitrary directory I get:
% gtk-config --libs --cflags --version
-L/usr/local/lib -L/usr/X11R6/lib -lgtk -lgdk -lglib -lXext -lX11 -lm
-I/usr/local/lib/glib/include -I/usr/local/include -I/usr/X11R6/include
