Yes.

As Jesse van den Kieboom mentioned in a previous mail, spawn a new gedit instance would reload the spec.

BTW you can put custom lang files in ~/.local/share/gtksourceview-2.

I would guess that adding your changed *,lang file to your home directory might cause a re-read when a new gedit document is opened especially if you use a command line "gedit newfile". I will try it right soon now.
