Adblock needs PCRE, maybe it's not installed (and beware, adblock will
crash on exit, that's why it's not enabled by default --- a mozilla
bug).

--with-extensions=really-all should have enabled all extensions (if
Epiphany was compiled with python support); can you show us the
config.log file that configuring epiphany-extensions produced?
