I think what you're after is set_default_response. E.g.:
my $file_chooser = Gtk2::FileChooserDialog->new(
'Open',
undef,
'open',
'gtk-cancel' => 'cancel',
'gtk-ok' => 'ok'
);
$file_chooser->set_default_response('ok');
